1. If you know the position of a promoter relative to the coding region of a gene, how can you tell which strand is the template for transcription? 

2. If you have a mutation that results in the deletion of the -10 sequence of an E. coli promoter, how would you expect transcription to be affected? 

3. If there is a mutation that results in the deletion of the start codon of a gene, how would you expect transcription to be affected?

a. Ligase 
b. Topoisomerase 
c. Helicase 
d. The active site in DNA polymerase that affects proofreading
