Ninhydrin (trikethydrindene hydrate) is a stron oxideIng agent which is able to carry out oxidative decarboxylation of amino acids to yield CO2, NH3 and an aldehyde and the reduced from ninhyderin . Reduced ninhydrin reacts with additional mole of ninhydrin in the presence of ammonia and gives a blue/ purple coloured products which is quite specific and shows absorption in the vicinity of 570 nm. Proline and hydroxylproline give a yellow color with ninhydrin. This reaction is useful for estimation of amino acids quantitatively with the help of spectrophotometer. One of the requirements of this reaction is to use two moles of ninhydrin solution with one mole of amino acid.
